Introduction Metabolic syndrome is becoming more prevalent, caused by immune dysfunctions and caloric excess. Recent data show protection from the ancient, helminthic-induced immunity, Type 2 immunity. Whole body vibration (WBV) is anti-inflammatory and reverses polyuria and polydipsia in T2DM. The objective of this study is to measure Innate Lymphoid Cells profile in blood and kidney before and after WBV. We hypothesize that WBV increases (% ILC-2)/ (% ILC 1 + % ILC-3), the “immune regulatory” index (IRI).
Methods 11 mice were used: 10 week-old male db/db (n=7) and db/m (n= 4). They underwent WBV (1+/-.5g, 20 Hz)x 20 minutes/day 7 days/week x 4 weeks. The peripheral blood and kidneys were assayed for ILC by FACS (T-bet, GATA-3, ROR-gt: ILC-1, 2, 3 respectively) and cytokines: ILC-1 (IFN-g/TNF-a), ILC-2 (IL-5/IL-13), and ILC-3 (IL-17/IL-22)
Results Before WBV, the blood showed 0.01%, 0.02%, 0% for ILC-1,2, and 3 in db/m. 0.02% 0.01%, 0.01% for db/db. Kidneys showed 0.02%, 0.05%, and 0.03% for db/m, 0.08%, 0.03%, and 0.07% for db/db. After WBV, the db/m blood ILCs showed no change, but had 0.03 % ILC-2 with no ILC-1, 3 in db/db. Post-WBV db/m kidneys showed 0.02%, 0.03%, 0.02% for ILC-1,2, 3 and 0.05%, 0.05%, 0.01% in db/db. Renal IRI showed 417% increase.
Conclusion WBV improves renal function in db/db mice with lower ILC-1, ILC-3, and more ILC-2, but not blood of db/db or blood and kidney of db/m. This may partially explain the WBV's anti-inflammatory effects, providing further support to the Immuno-modulatory capability of WBV.
