Several studies have shown that CD44 and CD74 are overexpressedand dysregulated in various type of cancer, including lung cancer, breast cancerand prostate cancer. There selective expression in tumours is known to activatesignalling mechanisms that drive tumour progression. CD44 is atransmembrane glycoprotein molecule that functions in cell proliferation,migration and adhesion and also acts as a receptor for hyaluronic acid. CD74is a non-polymorphic glycoprotein that functions in regulating the traffickingof MHC class II protein (Karakikes et al, 2012) and also serves as the receptorfor macrophage inhibitory factor (MIF). The MIF/CD74 receptor complex requiresCD44 as the signalling component. We have employedflow cytometry and to examine non-small cell lung cancer cell line, A549 forCD74 and CD44. We observed that CD44 was higher than CD74. UsingCo-localization experiments and bio imaging we performed quantitativevariations of CD74 and CD44 receptors in order to evaluate their association incytoplasmic compartments. We conclude that the two receptors CD44 and CD74 arenot colocalized in this cell line. This suggests that they modulate functionsas separate entities on non-small lung cell cancer.
