• The Echo 525 Liquid Handler enables a method to miniaturize the final reaction volume to prepare libraries using the Illumina® Nextera™ DNA library prep kit to 2.5 μL compared to the 50 μL volume
recommended by the manufacturer, for a 20-fold reduction in reaction volume.
• The implementation of a chemical clean-up and reduced reaction volume can significantly improve throughput and lower overall sequencing costs.
• Sample input, tagmentation volume, PCR reaction volume can be miniaturized using the Echo 525
liquid handler to increase throughput and reduce costs.
• Sequencing of the libraries prepared by these methods demonstrated equivalent quality metrics when compared to the manual reaction.
