Bivalves and other mollusks collected from the wild and from shellfish culture are susceptible to bacterial contamination from adjacent coastal and residential areas, and from algal blooms specially from toxin-producing species. The process of depuration aims to mitigate the presence of matters negatively contributing to food safety. In the study, two depuration techniques were evaluated based on their efficiency and the duration it takes for each method to significantly reduce contaminants specifically PST and coliform levels. Blood cockles and water samples obtained from Eastern Samar, Philippines were subjected to 24h and 48h depuration processes which consist of UV and ozone. Additionally, initial and 0h samples were also taken prior to depuration. The most probable number (MPN), total plate count, as well as biochemical tests through API 20 E kit for characterization and confirmation were performed. It was observed that depuration for 24h using UV better reduced the MPN of the shellfish as opposed to using ozone. However, at 48h, MPN value drastically decreased under the ozone treatment, lower than that of the UV. PST toxin level analysis was done using high performance liquid chromatography (HPLC).
